孕激素受体在人类心脏发育中具有重要作用

人心脏的发育从胚胎开始,一直持续到成年,以建立成熟的基因表达程序,从而在整个生命中维持心输出功能。尽管我们对控制胚胎心脏发育的分子机制已有深入的了解,但对控制人出生后心脏成熟的分子信号仍知之甚少。

近日,发表在《循环》“Circulation”上的一篇最新研究“Sex-Specific Control of Human Heart Maturation by the Progesterone Receptor”揭示了孕激素受体对人心脏成熟的性别特异性控制。

Choon等人对9位人类供体的54 140个细胞核进行了单核苷酸RNA测序,分析了各种类型的心脏细胞从胎儿期到成年期的转录变化。采用大量RNA测序和使用测序的转座酶可及染色质测定进一步在21位人类供体的纯化的心肌细胞核中验证了转录变化以及染色质可及性的整体变化。最后,研究人员还在人类多能干细胞衍生的心脏类器官和小鼠中进行了涉及心脏成熟的性类固醇的功能验证研究。

研究结果显示,在人类心脏发育过程中,存在年龄和性别依赖性的转录改变孕激素受体是心肌细胞成熟过程中性别依赖性转录程序的关键介体。在人类心脏发育过程中,心肌成熟与甾体激素核受体模体周围的性别特异性的染色质重塑有关。
