随着细胞疗法的日益发展,我们需要合适的祖细胞来源来减轻以及治疗疾病。近日,来自多伦多大学的研究人员提出了一个中断的重编程的策略。其在非增殖性的Club细胞中按照特定的时间点表达诱导多能性干细胞重编程因子(Oct4Sox2Klf4c-Myc; OSKM),以诱导产生的祖细胞样细胞。

中断的重编程允许控制细胞扩增,并且保留了其谱系承诺。在克隆形成条件下,iPL细胞扩增并且作为类细支气管祖细胞群发挥功能,以产生成熟的Club细胞,产粘蛋白的杯状细胞,和表达囊性纤维化跨膜传导调节剂(CFTR)的纤毛上皮细胞。

在体内,iPL细胞可以重新填充CFTR缺陷上皮。这种中断的重编程过程可能可以达到节制性的祖细胞样增殖的目的。

通过仔细控制OSKM表达的持续时间,iPL细胞不会变成多能性细胞,并且保持它们的起源记忆,保持其有效地恢复其原始表型的能力。

因此,这项产生高度特异性细胞的通用技术可能对再生医学产生重大影响。
