美国国家指南推荐采用低剂量计算机断层扫描对高危患者进行筛查,但这种方法存在局限性,包括假阳性率高。

活性纳米传感器可以检测体内失调的蛋白酶并释放报告基因,提供疾病活动的尿检读数。Kirkpatrick等人优化了基于蛋白酶活性的纳米传感器以筛查肺癌。

通过将纳米传感器与肺内递送和机器学习相结合,在两种具有免疫功能的小鼠模型中检测活性纳米传感器对肺癌的检测潜力。通过对人类和小鼠肺腺癌转录组数据集进行对比分析及使用重组候选蛋白酶进行的体外裂解测定,研究人员对传感器的多重面板进行设计。

通过对Kras和Trp53突变的肺腺癌小鼠模型肺内予以纳米传感器证实了金属蛋白酶在肺癌中的作用,并能以100%的特异性和81%的敏感性准确检测局部疾病。而且,在另一种肺腺癌自体模型中应用该方法时,检测出癌症的特异性达到了100%,敏感性达到了95%,且不受脂多糖驱动的肺部炎症的干扰。

本研究结果支持开发用于检测肺癌的活性纳米传感器。
