本研究通过引起猪的二尖瓣重度反流构建容量负荷型心衰模型,造模成功1个月后,将猪随机分成冠脉注射BNP116.I-1c(n=6)组或生理盐水组(n=7),随后,对其有效性和安全性随访评估2个月。此外,24只幼猪分别接受了不同剂量的BNP116.I-1c治疗以评估其安全性能。造模一个月后,猪均发展成为心衰,BNP116.I-1c治疗组猪的左室射血分数有所改善(5.9 ± 4.2% vs. 5.5 ± 4.0%; p < 0.001),并且在接受治疗2个月后,左房射血分数也有明显改善(4.3 ± 3.1% vs. 7.5 ± 3.1%; p = 0.02)。在体外,猪或大鼠的左房细胞在暴露于I-1c后表现出明显的钙离子电流幅度增大。在BNP116.I-1c治疗组中,没有发现明显的不良心律失常、细胞免疫应答激活和非靶器官的损害。

研究结果表明,冠脉给予BNP116.I-1c治疗可以明显提高心衰模型动物的左室和左房的收缩功能。
