Ji-Xin Cheng早先带领研究团队发现前列腺癌的侵袭性与细胞内胆固醇代谢产生的一种化合物之间存在关联,在这项发表在国际学术期刊Oncogene上的最新研究中,他们将这一发现进一步拓展到胰腺癌,同时表明一类之前开发用于治疗动脉粥样硬化的药物或有希望用于胰腺癌和其他癌症的治疗。

研究人员发现在人类胰腺癌样本和细胞系中存在胆固醇酯的积累,表明胆固醇的酯化与癌细胞转移之间可能存在关联。酯化是胆固醇在细胞内储存所需要的一个生化过程。过量胆固醇会导致胆固醇以胆固醇酯的形式存在于癌细胞内部的脂滴中。抑制胆固醇酯的形成或可成为治疗转移性胰腺癌的一种新策略。

研究人员开发了一种叫做Raman spectromicroscopy的分析工具对活细胞内的单个脂滴进行成分分析。结果发现胆固醇酯在人类胰腺癌样本和细胞系中存在异常积累,抑制酯化过程能够显著抑制小鼠体内胰腺肿瘤的生长和转移。随后研究人员用一种抑制酯化过程的动脉粥样硬化治疗药物——avasimibe对肿瘤原位移植小鼠模型进行了药物治疗效果检测,avasimibe是一种叫做ACAT-1的酶的抑制剂,胆固醇酯的积累受到ACAT-1酶的控制,ACAT-1表达越高,病人生存几率越差。结果表明avasimibe能够减少脂滴数目,Raman分析结果也证实脂滴内部的胆固醇酯也出现减少,这表明avasimibe能够阻断胆固醇酯化过程,抑制肿瘤生长和转移。除此之外,阻断胆固醇酯的储存还会引起内质网损伤,导致癌细胞死亡。

这项研究表明靶向癌细胞内的胆固醇代谢过程或可帮助抑制肿瘤生长和转移,对未来开发靶向治疗方法提供了重要信息。
