RNA干扰是基因表达转录后调控的主要机制。microRNA(miRNA)前体被转录,加工成成熟的miRNA,并加载到Argonaute(AGO1-4)蛋白上,形成RNA诱导的沉默复合物RISC。因此,通过RNA干扰精确调控基因的表达对于正确开发和维持复杂的神经回路至关重要。

既往研究显示,AGO2(ARGONAUTE-2)和相关的miRNA形成RNA诱导的沉默复合物(RISC)能够通过靶向mRNA介导翻译沉默和降解,成为RNA干扰的一部分。Ago2的双等位基因缺失也会导致小鼠表现出包括中枢神经系统异常在内的各种发育缺陷,进而导致早期胚胎的致死。然而,目前对于任何人类病理学相关的编码AGO2基因的遗传改变尚不清楚,且RISC在人类发育和器官功能中的作用也知之甚少。

AGO2突变的鉴定

研究人员鉴定了21名受神经发育障碍影响的患者中AGO2基因的13个杂合突变。而每个鉴定出的单氨基酸突变均会导致shRNA介导的沉默受损。而作为突变引起的特异性功能改变,研究人员观察到了RISC的形成受损或AGO2与mRNA靶标结合的增加。后者主要是通过mRNA靶标释放的C端丝氨酸蛋白簇的磷酸化水平降低、神经元中树突状P小体的增加以及患者来源的原代成纤维细胞的整体转录组改变所引起的。

综上,该研究结果揭示了动态AGO2-RNA关联对于人类神经元发育过程中基因表达调控的重要性。
