Cell Research:破解肝细胞体外培养和扩增难题

众所周知,肝脏在机械损伤后具有很强的再生增殖能力。最近报道,肝损伤后肝细胞补充也可以通过发生在肝细胞和duct-like祖细胞之间的可逆转变来实现。

近日,来自我国第二军医大学王红阳院士团队在线发表了关于肝细胞体外培养和扩增的研究,相关研究成果刊登于国际杂志Cell Research上。

众所周知,肝脏在机械损伤后具有很强的再生增殖能力。最近报道,肝损伤后肝细胞补充也可以通过发生在肝细胞和duct-like祖细胞之间的可逆转变来实现。肝细胞在体内具有强大的再生潜能,正常肝脏三分之二切除后可在一周内通过细胞增殖恢复原来肝脏体积。然而,原代肝细胞在体外培养条件下却无法实现体外长期增殖,这也一直是困扰该领域的瓶颈问题。解决这个问题有助于推进肝细胞体外药物代谢,终末期肝病细胞治疗以及疾病模型构建等研究。

研究人员在前期研究发现,肝脏在特定损伤环境下,部分肝细胞可向肝前体细胞可逆转变,在躲避损伤的同时获得快速增殖的能力,待损伤因素减轻后可重新分化为成熟肝细胞来维持肝脏的基本功能。这提示如果能在体外模拟肝损伤环境下肝细胞信号通路的变化便有可能实现肝细胞的体外增殖。

依据这一理论,研究团队分别成功地实现了原代肝细胞在体外向肝前体细胞的转变和快速增殖,经定向诱导分化后,增殖的前体样肝细胞可重新获得成熟肝细胞的功能。
