尽管医学界一致认同适量运动有益心脏健康,但其原因至今尚未完全明确。例如,目前已知心肌细胞消失是导致心力衰竭的主要原因之一,并且成年人群心脏的心肌细胞生成能力非常有限(年轻成人每年仅约更新1%的心肌细胞,随着年龄增长会变得更少),但究竟是何种因素在对其进行调节,以及能否有效控制几乎都一无所知。在当前这项研究中,美国哈佛大学干细胞与再生医学系的Ana Vujic博士及其同事评估了运动这种安全且廉价的干预方法对成年小鼠心肌细胞再生能力的影响。

该项研究中,一组健康小鼠被放置在跑步机上,每天自由跑动约5km;另一组小鼠则没有进行此类运动,使其保持久坐状态。为了评估小鼠的心肌再生能力,研究者使用了一种标记后的化学物质,当心肌细胞准备分裂时,能够结合进入新生成的DNA中。通过追踪心肌中这些标记后的化学物质,可以观察到新细胞的生成情况。此外,研究者还采用同样方法评估了运动对心肌梗死模型小鼠的影响。

结果显示,经过8周的运动训练后,可以使成年小鼠新心肌细胞的生成大幅升高(相比于不运动小鼠升高约4.6倍);而在心肌梗死模型小鼠中,运动在梗死区域的广泛边界部分诱导产生了极强的心肌生成应答。

该研究的两位资深作者Richard Lee博士(哈佛大学干细胞与再生生物学教授)和Anthony Rosenzweig博士(哈佛干细胞研究所主要成员)共同指明两点:

1.为了维持健康心脏,需要通过心肌细胞再生或生成新的心肌细胞来平衡损伤或老龄因素造成的心肌细胞丢失。当前研究结果提示,运动有助于实现这一有利于心肌细胞再生的平衡关系,每天运动或许可以使心脏变得更加年轻。”

2.当前这项研究确定了运动启动心肌细胞再生所需的具体生物学通路,现在,我们需要进一步发现足以开启这一通路的信号。如果我们能够在恰当的时间和患者中启动这些通路,就有望改善患者心脏病发作后的恢复情况。”
