【1】MiR-218可促进骨髓间充质干细胞的成骨分化并加速骨折愈合

作为成骨调节剂,研究报道microRNA-218(miR-218)可促进间充质干细胞(MSCs)的成骨。 然而,miR-218的体内成骨作用尚未清楚。在这项研究中,研究人员发现miR-218通过刺激碱性磷酸酶活性,钙结节形成和成骨标记基因表达来证实能促进MSCs的成骨分化。体内研究中,将miR-218过表达的BMSCs局部施用于股骨骨折小鼠模型的骨折部位。X射线、微型计算机断层扫描、力学测试、组织学和免疫组化检查显示,miR-218过表达改善了新骨形成并加速了骨折愈合。表明未来miR-218可能成为临床应用中骨修复的有希望的治疗靶点。

【2】miR-185抑制肝星状细胞激活和纤维化进展


最近的研究表明,microRNAs对肝星状细胞(HSC)的激活和转化产生影响,这对于肝纤维化病理机制的研究有重大意义。本研究旨在探究miR-185在肝纤维化中发挥的作用。结果发现健康对照(n=8)和HBV相关肝纤维化患者(S2/3,n=10)的血浆中可检测到miR-185,肝纤维化患者中水平显著下调。在ccl4诱导的肝纤维化小鼠肝组织中和TGF-beta1激活的HSCs中同时也存在miR-185水平的下调,同时RHEB和RICTOR的表达增加。将LX-2细胞转染了miR-185后发现,alpha-SMA、胶原1和胶原III水平下降,RHEB和RICTOR的表达也下降。内源性的miR-185的抑制,导致肝纤维化活性的增强。双荧光素酶报告检测表明,miR-185抑制了RHEB和RICTOR的表达。此外,RHEB和RICTOR的沉迷抑制了a-SMA和胶原的表达。表明miR-185通过抑制RHEB和RICTOR的表达进而抑制HSC的激活和肝纤维化的进展。

【3】循环miR-32水平改善慢性心力衰竭患者
心衰住院风险预测

近日,研究人员在慢性心衰患者中进行了一项多中心临床试验分析了特定候选miRNA的预后价值。使用miRNA特异PCR技术检测了953例慢性有症状心衰患者血浆中miR-32水平。在校正模型中,研究人员估计了致命性(全因和心血管死亡)和非致命事件(心血管原因或心衰恶化的首次入院时间)与增量风险预测之间的相关性。结果发现较高的循环miR-32水平与年龄较小的、肾过滤较好的、缺血性病因的HF、症状更严重HF、舒张压高、高胆固醇和男性独立相关。校正混杂因素后发现,miR-132水平只与心衰住院相关,并可以改善其风险预测。由此可见,在典型的慢性心衰患者中,循环miR-32水平升高与心衰的严重程度相关。低水平循环miR-32相比于传统的危险因素,可提高心衰再入院的风险预测,但不能提高对死亡的预测。miR-32可能有助于强化旨在减少再住院的策略。

【4】LncRNA XIST通过miR-195-5p靶向YAP促进骨肉瘤进展


lncRNA XIST(X非活性特异性转录物)是存在于各种恶性肿瘤中的致癌lncRNA,然而,其在骨肉瘤(OS)发展中的作用和分子机制尚不清楚。本研究中,研究人员收集了40对OS组织和匹配的相邻非肿瘤组织,发现OS组织和细胞系中XIST的表达均明显上调,与患者的临床预后呈负相关。敲除XIST可抑制体外癌细胞的增殖和侵袭,抑制体外OS细胞的EMT,并抑制体内皮下肿瘤的生长。进一步的分析表明,XIST通过在OS细胞中作为竞争性内源RNA海绵miR-195-5p来调节YAP的表达。 XIST直接与miR-195-5p相互作用,降低了miR-195-5p与YAP 3'UTR的结合,抑制miR-195-5p对YAP mRNA的降解。表明lncRNA XIST部分通过miR-195-5p/YAP通路增强OS癌细胞的增殖和侵袭。因此,lncRNA XIST可能是O治疗的靶标。

【5】miR-181a-5p 和 miR-23a-3p表达变化与肥胖及胰岛素抵抗有关

虽然一些miRNA被认为与脂肪组织(AT)IR的发展有关,但是miRNA在炎症与IR之间的关联中的作用尚不清楚。Javier Lozano-Bartolomé等人则研究了miR-181a-5p和miR-23a-3p在肥胖中的表达谱并研究它们脂肪细胞中TNFα诱导的IR中作用。结果显示,来自肥胖和糖尿病受试者的AT中miR-181a-5p和miR-23a-3p的表达降低,并且与肥胖和HOMA-IR呈负相关。在脂肪细胞中miR-181a-5p和miR-23a-3p的过度表达上调了胰岛素刺激的AKT活化并降低了TNFα诱导的IR,从而调节PTEN和S6K表达。与对照组相比,病例组基线水平血清miR-181a-5p水平降低,指向其预后价值。投影评分变量的重要性表明,miR-181a-5p在120分钟时比胰岛素或葡萄糖对模型的影响更大。表明miR-181a-5p和miR-23a-3p可以通过调节PTEN和S6K的表达来阻止TNFα诱导的脂肪细胞的IR。
