用小分子RNA疗法治疗小鼠急性骨髓性白血病

急性骨髓性白血病是一种血细胞癌症,往往在进行了密集化疗后依然会在5年内夺走患者的生命。虽然当前已有许多关于急性骨髓性白血病基因组变异的特征信息,但人们对驱动祖细胞转化成癌症细胞的分子机制却所知甚少。

美国辛辛那提大学陈建军团队分析了62名患者的癌症样本,发现急性骨髓性白血病患者的miR-22表达减少了。研究使用急性骨髓白血病的多种小鼠模型发现,可以通过抑制特定的细胞传导通路恢复miR-22的表达水平,阻碍急性骨髓性白血病的发展和癌细胞的维持。研究人员随后通过采用纳米颗粒运送miR-22短链RNA到两种白血病小鼠模型评估miR-22疗法的效应。在两种模型中,使用miR-22治疗都带来了癌症进展的延迟与生存时间的延长。

不过,研究人员指出,在对携带miR-22的纳米粒子治疗急性骨髓性白血病进行临床试验之前,还需要进一步的测试。他们还表示,把这种疗法和标准的化疗药物相结合也能有效治疗急性骨髓性白血病
