近日,《Blood》上最新发表的“ER associated degradation preserves hematopoietic stem cell quiescence and self-renewal by restricting mTOR activity”报道:通过内质网相关降解(ERAD)控制的蛋白质质量,调控静止造血干细胞(HSCs)的功能。

Sel1L/Hrd1 ERAD基因在静止和不活跃的造血干细胞中富集

在静止和不活跃的造血干细胞中,Sel1L/Hrd1 ERAD基因富集,而条件性敲除造血组织中的Sel1L可导致HSC过度增殖,导致HSC完全丧失自我更新能力,最终导致HSC耗竭。

Sel1L缺失导致HSC过度增殖和激活

在机制上,通过敲除Sel1L基因导致ERAD缺陷,进而使mTOR信号激活。此外,研究人员还发现mTOR激活剂Rheb是Sel1L/Hrd1 ERAD的一种新的蛋白底物,它在Sel1L缺失和HSC激活时积聚。

重要的是,抑制mTOR或Rheb可以挽救Sel1L基因敲除小鼠的HSC缺陷。

综上所述,通过ERAD的蛋白质质量控制是一个关键的检查点,它通过Rheb介的mTOR活性限制来调控HSC的静止和自我更新。
