Sci Rep:在怀孕时吸烟竟能增加隔代子女患自闭症风险!

自闭症和自闭症谱系障碍使儿童缺乏像其他孩子那样能与世界互动的能力,自闭症谱系障碍(ASD)的特征是社会互动困难,沟通存在问题和习惯从事重复行为。虽然自闭症谱系障碍的遗传背景有很多的研究,但环境因素有可能导致患病率随时间的变化。啮齿动物实验表明,环境暴露可以对后代产生影响;人类研究表明,父母产前暴露可能在发育变化中发挥重要作用。

自闭症和自闭症谱系障碍使儿童缺乏像其他孩子那样能与世界互动的能力,自闭症谱系障碍(ASD)的特征是社会互动困难,沟通存在问题和习惯从事重复行为。虽然自闭症谱系障碍的遗传背景有很多的研究,但环境因素有可能导致患病率随时间的变化。啮齿动物实验表明,环境暴露可以对后代产生影响;人类研究表明,父母产前暴露可能在发育变化中发挥重要作用。

这项研究使用父母和儿童的Avon纵向研究(ALSPAC)来验证假设,如果母亲或父亲(F1)在怀孕期间暴露于自己的母亲(F0)吸烟,则后代(F2)患自闭症的风险将增加。我们发现在怀孕期间外祖母吸烟与在社会交往和重复行为措施中有不利评分的孙女(外孙女)之间的联系可以独立预测诊断自闭症。根据这一点,我们将吸烟外祖母的外孙子/女中的自闭症的实际诊断相关联。在怀孕期间祖母吸烟和孙子女自闭症并没有关联。

利用父母对子宫内吸烟进行的代际影响研究有两个原因。首先这是一种容易记忆的习惯;第二,已知妊娠吸烟对儿童发育和DNA甲基化有不利影响,而对下一代的影响在理论上是可能的。进行了分析,以确定自闭症状和子宫内父母接触吸烟之间是否存在性别特异性关联,但不能确定具体是祖母或外祖母或儿童的性别之间的具体联系。因此,在其他研究中证实这些联系是特别重要的。
