Laurel E. Hind等人开发了一种微型的包含内皮细胞腔、3D细胞外基质和完整的细胞来源的三维(3D)模型来模拟宿主微环境。通过该模型,研究人员发现内皮细胞腔可显著增加中性粒细胞向绿脓杆菌的源头迁移。中性粒细胞一直以来被认为在体外时存活时间短,但研究人员发现在内皮细胞腔和细菌存在的情况下,中性粒细胞在3D缓解中迁移可长达24小时。此外,研究人员还发现内皮细胞在绿脓杆菌的诱导下可分泌炎症介质,包括GM-CSF(可促进中性粒细胞存活)和IL-6(一种促炎症细胞因子)。

研究人员还发现用抗TL-6受体的封闭抗体预处理中性粒细胞可显著减少向绿脓杆菌迁移的中性粒细胞,但不影响中性粒细胞的存活时间,提示分泌的IL-6是内皮细胞和中性粒细胞之间的重要信号,可调节中性粒细胞迁移。

总而言之,本研究表明内皮细胞的旁分泌信号参与调控中性粒细胞的迁移和存活。
