来自阿尔伯塔大学的研究人员开发出了一种使用光控制生物细胞水平的新方法。这种工具被称为光可切割蛋白(photocleavable protein):PhoCl,也就是当暴露于光线时,蛋白会裂解开来,这样科学家们就能以新的方式来研究和操纵细胞内活性。

来自阿尔伯塔大学的研究人员开发出了一种使用光控制生物细胞水平的新方法。这种工具被称为光可切割蛋白(photocleavable protein):PhoCl,也就是当暴露于光线时,蛋白会裂解开来,这样科学家们就能以新的方式来研究和操纵细胞内活性。

这一研究成果公布在3月的Nature Methods杂志上,领导这一研究的是阿尔伯塔大学的蛋白质工程师Robert Campbell。他曾说过,“在我的职业生涯中期阶段,我发现要想单纯的进行发明有些可怕。由假说驱使的研究总是会得到一个答案,无论得到的是不是正确的答案。相比之下,由发明驱动的研究往往有双向出口,要不成功,要不失败,而失败很少有人提及。”Nature技术人物:想要发明,而不是验证的科学家

Campbell师承加州大学圣地亚哥分校的华裔诺奖得主Roger Tsien(钱永健)。他在自己的职业生涯中研发出了一种称为FPX的新技术工具,这种工具能利用荧光蛋白在活细胞和组织里成像动态的生化事件,将蛋白互作的变化转变为立即可见的颜色改变(红-绿),这不仅为人们提供了即时检测细胞变化的新工具,还向人们展示了构建新一代生物感应器的一种通用策略。

在这篇文章中,研究人员首先利用光可切割蛋白将细胞蛋白与抑制剂连接在一起,令其无法执行正常的功能,这个过程被称为锁定。Campbell表示,“(然后)通过将光照射到细胞中,我们可以使光可切割蛋白裂解,从而去除抑制剂,令细胞内蛋白解锁”,一旦蛋白被解锁,那么就可完成其在细胞内的正常功能。

这一工具对于希望能调控细胞内进程的研究人员来说,相对容易使用,也应用广泛。

Campbell解释说,光敏蛋白可以被用于研究任何活细胞的内部工作。例如,光遗传学工具广泛应用于激活小鼠的脑活动。“我们可以利用光可切割蛋白在实验室研究单个细菌,酵母,人类细胞,甚至整个动物,如斑马鱼或小鼠。不过首先需要将这些蛋白放入动物体内,因此我们简单地将这种蛋白的基因剪接成DNA,利用已有技术将其插入到细胞中。”
目前Addgene可以提供光可切割蛋白基因。

“我们希望提供新的方法来了解细胞生物学,”Campbell说,“未来这将能有更多潜在的应用,比如研究哪些细胞会成为发育生物学中的哪些组织,以及分析基因编辑技术的可行性。”

光遗传学是近年来最具创新性的显微技术之一,通过结合遗传学和光学方法,科学家们可以利用光来特异性控制活细胞中精确时间段的蛋白活性,以及蛋白相互作用。

在进行光遗传学实验的时候,研究人员经常需要使用一种激光共焦显微镜的光漂白模式(photobleaching mode)。虽然目前特殊激光或其它通过光学半导体作为数字微镜设备(digital micromirror devices)的照明系统,已经得到了长足发展,但是这些设备价格依然居高不下,而且也只能用于细胞生物学研究中的几种光应答蛋白。此前来自日本香川大学的研究人员研发出了一种自动荧光显微系统,这种系统采用传统的光学零件和软件,能操控活细胞中的基因编码光应答蛋白。自动荧光显微系统:高效光控蛋白。
