既往范可尼贫血(FA)的相关研究均未涉及FA患者的基因校正后的造血干细胞和祖细胞(HSPC)的移植效果,不管是自体移植后还是输注到免疫缺陷小鼠。

在本研究中,研究人员证明G-CSF联合plerixafor借助治疗用的FANCA-慢病毒载体诱导FA-A患者的CD34+细胞短期转化后,可校正体外培养的造血祖细胞的表型。向免疫缺陷小鼠输注转化的FA CD34+细胞,可使骨髓、淋巴和CD34+细胞再生。重要的是,移植后,在注入CD34+细胞移植物中可观察到表型校正的患者来源的造血细胞的比例显着增加,表明注入的校正后的FA-A细胞具有增殖优势。研究数据首次证明了HSC系列的优化方案,用治疗性的慢病毒载体对这些细胞进行的短期且已临床验证的转化,可获得能重回输并在免疫缺陷小鼠体内具有增殖优势的表现校正了的HSPCs。
