Blood:从微流体看血小板和活化内皮的相互作用。

本综述主要是基于体外流式实验和体内血栓形成实验的交叉参考对血小板-内皮界面进行阐述。

一直以来,均认为体外流式实验和体内血栓形成研究对阐述血管壁受损后血小板血栓形成机制至关重要。近年来,逐渐发现血小板还在炎症过程中发挥调节作用,如动脉粥样硬化。其中一个关键元素是血小板、凝血系统、白细胞和活化内皮之间的复杂的交联。

本综述基于体外流式实验和体内血栓形成实验的交叉参考对血小板-内皮界面进行阐述。

血小板和活化的内皮细胞之间的相互作用机制:

1)血小板通过血小板糖蛋白Ib-IX-V与内皮细胞释放的血管性血友病因子相互作用而发生滚动,P选择素与P选择素糖蛋白配体相结合;

2)血小板通过血小板αIIbβ3与内皮细胞αvβ3和细胞间黏附分子l结合而黏附到血管内皮;

3)凝血酶、血小板反应蛋白-1-CD36轴和环氧化酶1在随后的血小板活化和稳定血栓形成过程中发挥了激活作用。

此外,本文还讨论了血小板对白细胞迁移(动脉粥样硬化形成过程的关键过程)的刺激作用的分子机制。
