本周《自然》在线发表的一项研究Quantitative phosphoproteomic analysis of the molecular substrates of sleep need表明,大脑中的蛋白质磷酸化水平可能驱动着睡眠欲望。该研究揭示了睡眠需求的分子基础,并强调了与睡眠有关的疗法的潜在分子靶点。

本周《自然》在线发表的一项研究Quantitative phosphoproteomic analysis of the molecular substrates of sleep need表明,大脑中的蛋白质磷酸化水平可能驱动着睡眠欲望。该研究揭示了睡眠需求的分子基础,并强调了与睡眠有关的疗法的潜在分子靶点。



嗜睡突变小鼠的大脑表现出过度磷酸化,模拟睡眠剥夺小鼠大脑中的磷酸化。

在哺乳动物中,睡眠-觉醒周期由平衡睡眠需求和实际睡眠时间的体内平衡机制驱动。美国德克萨斯大学西南医学中心的Qinghua Liu及其同事研究了睡眠剥夺和嗜睡(Sleepy)突变小鼠模型的脑蛋白磷酸化水平,发现总体磷酸化水平与睡眠需求相关。睡眠降低磷酸化水平,而长时间觉醒导致过度磷酸化和高睡眠需求。



SLEEPY(变异的SIK3蛋白)与睡眠需求指数磷蛋白相互作用,改变睡眠-觉醒稳态。

作者鉴定了80个主要为突触蛋白的蛋白,其磷酸化状态根据睡眠需要而发生改变。这引人深思,因为突触可塑性也与睡眠有关。根据突触平衡假说,睡眠使突触有机会从其日常活动中恢复过来,维持平衡状态。因此,突触蛋白的磷酸化可能是维持突触平衡和睡眠-觉醒平衡的关键。
