J Clin Invest:PD-L1在GVL与GVHD中的研究进展

目前,由于对供体衍生的同种异体反应性T细胞介导的移植物抗白血病/淋巴瘤(GVL)作用的良好认识,异基因造血细胞移植(HCT)代表了可用于治疗各种血液恶性肿瘤的一种方法。然而,移植物抗宿主病(GVHD)是由相同的T细胞介导,并且其仍然是导致大量发病率和死亡率的重大临床问题。近期,在杂志JCI发表的文章中,研究者Ni及其同事使用了几种GVHD鼠模型来评估CD4 + T细胞的消耗对GVL与GVHD的影响


目前,由于对供体衍生的同种异体反应性T细胞介导的移植物抗白血病/淋巴瘤(GVL)作用的良好认识,异基因造血细胞移植(HCT)代表了可用于治疗各种血液恶性肿瘤的一种方法。

然而,移植物抗宿主病(GVHD)是由相同的T细胞介导,并且其仍然是导致大量发病率和死亡率的重大临床问题。

近期,在杂志JCI发表的文章中,研究者Ni及其同事使用了几种GVHD鼠模型来评估CD4 + T细胞的消耗对GVL与GVHD的影响,并显示了CD4 + T细胞的消耗导致通过受体组织和供体CD8 + T细胞介导的PD-L1的上调。 

PD-L1与PD-1在GVHD靶向组织中的相互作用导致CD8 + T细胞的衰竭和凋亡,从而阻止GVHD的发生,而PD-L1与淋巴组织中CD80的相互作用促进CD8 + T细胞的存活和扩增,从而增强了GVL反应。

这些研究结果可能为开发有效的临床策略以增强GVL,同时最小化同种异体HCT后发生GVHD奠定基础。
